BIOSENSORS Extraction and Detection of Pesticides by Cholinesterase Inhibition in a Two-Phase System: a Strategy to Avoid Heavy Metal Interference

چکیده
A novel method of extraction has been developed to avoid the presence of heavy metals during the measurement of pesticides based on acetylcholinesterase (AchE) inhibition. Heavy metals have been in fact demonstrated in this article to interfere when the assay is performed by using the classic spectrophotometric Ellman’s method. We present the results obtained with an assay system using two different phases, one organic and the other aqueous, in which the pesticide and the enzyme are, respectively, solubilized. In a first step, the concentration of the substrate acetylthiocholine (1 mM), of the enzyme (7 mU mL), and the reaction time (20 min) for measurement of enzyme activity were optimized in aqueous solution. Next, the effect of an organic phase on the enzyme activity was studied by the addition of various solvents with the activity being evaluated after 10 min of mixing. It was found that by using hexane, the enzyme retained almost 100% of its Received 18 April 2005; accepted 15 May 2005 The authors thank the MIUR project FISR-1999 and the EU project BIOCOP for

A conductometric biosensor using immobilised Chlorella vulgaris microalgae as bioreceptors was used as a bi-enzymatic biosensor. Algae were immobilised inside bovine serum albumin membranes reticulated with glutaraldehyde vapours deposited on interdigitated conductometric electrodes. Local conductivity variations caused by algae alkaline phosphatase and acetylcholinesterase activities could be ...

Phosalone, malathion and diazinon were analyzed in corn oil using solid phase extraction (SPE)with lanthanum silicate as a new solid sorbent followed by gas chromatography with nitrogenphosphorus detection (GC-NPD). The efficiency of different eluents (methanol, dichloromethane, ethyl acetate, dichloromethane — ethyl acetate (1:1) and ethyl acetate — methanol (1:1)) werecompared for elution of o...

This article presents the method of extraction-preconcentration of Lead, Cadmium, and Nickel ions from food samples using the Ghezeljeh montmorillonite nanoclay (Geleh-Sar-Shoor) as a new native adsorbent in batch single component systems. The extraction-preconcentration of heavy metals were carried out by applying the solid phase extraction (SPE) method followed by atomic abs...

Butyryl cholinesterase of different origin along with variations of the time of enzyme immobilization on the potentiometric transducer surface is offered to control the ion sensitive field effect transistor (ISFET)-based biosensor sensitivity. Because butyryl cholinesterase has been already used to develop the sensors for heavy metals, organophosphorus/carbamate pesticides, and steroidal glycoa...
